We are coming from Rome by train. Is the departure point near this train station where we will arrive ?

Probably not. I recommend the itaxi app for transport from the train to the departure point.
Q:
Is this tour recommended for a couple traveling with a baby (1 year old) ?
A:
The whole day is about 12 hours, with 5 - 6 hours of travel time in a bus (with no restroom). All sites are stroller friendly. Probably not the best tour to take with a 1 yr old.

We will have 2 nights and one full day there. Can anyone suggest a reliable tour to explore Val D'orcia, Pienza, Montalcino or other breathtaking places in southern part of Tuscamy in that one day? Later, we will head to Siena and Florence.
A:
Hi! I went to Tuscany in one day tour from Florence, it was great and it will take all day but you will see lots of Tuscany, Siena, San Gimilano, and a wonderful farm.
